Our data on LYP phosphorylation agrees with a recent report [14], although there are discrepancies, for example in the kinetics of LYP phosphorylation, which seems delayed in our assays, probably due to the different cell lines used in each case. The major sites phosphorylated by LCK appeared to be Tyr526 and Tyr536. However, it remains elusive the role of Tyr phosphorylation for LYP function in TCR signaling, as mutation of several Tyr to Phe, including Tyr526 and Tyr536, did not alter the negative regulatory role of LYP in TCR signaling.
